提高膜片钳实验成功率的方法研究

Research on the Method of Improving the Success Rate of Patch Clamp Experiment

  • 摘要: 为了研究提高膜片钳实验成功率的方法,该文以大鼠肾上腺髓质嗜铬瘤分化细胞(pc12)的全细胞膜片钳电生理检测实验为例,通过分析实验影响因素和控制关键参数,记录高阻封接、细胞破膜成功率及质量,经实验数据分析,最终确定实验的最优参数。实验表明,在该文实验设备及环境下,提高成功率的最优参数为:玻璃微极管尖端直径为3 μm;Ag/AgCl电极电镀电流为0.1 mA,电镀时间为2 h;高阻封接负压在1~2 mL间;破膜负压在0.5~1 mL间;破膜电击刺激延续脉冲为3 ms。

     

    Abstract: In order to study the method of improving the success rate of patch clamp experiment, taking the whole cell patch clamp electrophysiological detection experiment of pc12 cells as an example, by analyzing the influencing factors and controlling the key parameters of the experiment, the success rate and quality of high blocking sealing and cell membrane breakage were recorded, and the optimal parameters of the experiment were finally determined through the analysis of experimental data. The experimental results show that under the experimental equipment and environment, the optimal parameters to increase the success rate are: the tip diameter of glass microelectrode is 3 μm; the plating current of Ag/AgCl electrode is 0.1 mA and the plating time is 2 hours; the negative pressure of high resistance sealing is 1~2 mL; the negative pressure of film breaking is 0.5~1 mL; and the continuation pulse of film breaking electric shock stimulation is 3 ms.
