超高通量测序模板的质控评估方法研究

Study of Quality Assessment of Ion Sphere Particles for Ultra-high Throughput Sequencing

  • 摘要: Ion Torrent作为革新性的芯片测序技术,具有简捷、快速、高效等优势,在生命科学研究领域展现出强大的能力。主要操作步骤包括:准备文库、制备模板、芯片测序和数据分析。其中,制备出含DNA链的离子微球颗粒的测序模板是试验的关键步骤之一。对离子微球颗粒进行流式细胞检测,可以在下一步测序试验进行之前,对离子微球颗粒制备情况作质控评估,减少因测序模板制备不成功或含DNA链的离子微球颗粒比例少所损耗的时间和试剂成本,提高测序效率。

     

    Abstract: Ion Torrent, as an innovative chip sequencing technology, has the advantages of simplicity, speediness and efficiency. It shows great ability in the field of life science research. The main steps include: preparation of the library, preparation of the ion sphere particles (ISPs) of the template-positive ion PGMTM template, chip sequencing and data analysis. One of the key steps of the experiment is to prepare the ISPs of the template-positive ion PGMTM template. In this study, flow cytometry is used to evaluate the quality control of enriched ISPs before the next step of the sequencing experiment, so as to reduce the cost of time and reagent caused by the unsuccessful preparation of ISPs, and improve the efficiency of sequencing.
