氮杂碳量子点的合成、表征及生物成像性能研究的综合实验设计

Comprehensive Experiment on Synthesis, Characterization and Bioimaging Performance of N-doped Carbon Dots

  • 摘要: 针对生物探针光学稳定性和生物相容性的要求,设计了氮掺杂碳量子点(N-CDs)合成、表征及生物成像性能研究的综合性实验。实验以具有良好生物安全性的山药粉和乙二胺为原材料,采用一步水热法制备了氮掺杂碳量子点。通过透射电子显微镜、X射线光电子能谱、红外光谱、紫外及荧光光谱对N-CDs的形貌、结构和光学性能进行表征和分析,并通过细胞毒性、生物成像实验考察了N-CDs的生物相容性和体内、外成像效果。该实验涵盖了材料制备、结构表征、生物相容性评价、生物成像等研究内容,系统展现了科学研究的完整过程,通过实验有助于培养学生的基础科学思维,提升学生的实践创新能力。

     

    Abstract: For the requirements of optical stability and biocompatibility of bioimaging probe, a comprehensive experiment on the synthesis, characterization and biological imaging performance of nitrogen-doped carbon quantum dots (N-CDs) was designed. Firstly, N-CDs was synthesized by a facile one-step hydrothermal approach using yam powder and ethylenediamine as raw material. Then, the morphology, structure and and optical property of N-CDs were characterized and analyzed by transmission electron microscopy (TEM), X ray photoelectron spectrometry (XRP), Fourier Transform infrared spectroscopy (FT-IR) spectrometry, UV-Vis and fluorescence spectrometry. Moreover, the biocompatibility and bioimaging performance of N-CDs in vivo and in vitro were investigated by cytotoxicity, cell imaging and zebrafish imaging analysis. The experiment involved integrated contents such as the novel materials preparation, structural characterization, biocompatibility evaluation and bioimaging, which exhibited the complete process of scientific research. Through the experiment, it is helpful to cultivate students’ basic scientific thinking and improve students’ practical innovation ability.
