针对代次较多的3T3-L1细胞诱导分化法的改良

Modification of the Inducing Method for the Differentiation of 3T3-L1 Cells with High Frequency of Doubling Towards Mature Adipocytes

  • 摘要: 小鼠前脂肪细胞3T3-L1被广泛应用于相关脂肪代谢、肥胖、糖尿病等的研究,诱导其分化为成熟脂肪细胞成为研究起点,但不少实验室采用经典诱导法时遭遇失败。为此改良经典诱导法,以提高分化成功率。首先,启动诱导的时机改变在细胞汇合度达到70的时候,而不是达到100再融合2d的时候;然后,对细胞孔进行前期的明胶处理。结果表明,采用改良方法后,分化成功率大幅提高。改良后的方法至少适用于传代次数约在10代左右的3T3-L1细胞。

     

    Abstract: The murine pre-adipocyte cell 3T3-L1 is widely used in the research related to the fat metabolism,obesity and diabetes etc,in which the successful induced differentiation of 3T3-T1 to mature adipocytes is the start point,but many laboratories are encountered the failure of induced differentiation with the classical induction method.To modify the classical induction method to increase the success rate,particularly towards the 3T3-L1 with around 10 times of doubling.Firstly,the start point to initiate the differentiation was set at the 70 confluence rather than 2 days after 100 confluence;Secondly,cell wells were pre-treated with gelatin.The modified method increased the success rate greatly.The modified method was at least suitable for the 3T3-L1 with around 10 times of doubling.
