Abstract: The conventional microscopic section making technique plays an important role in biomedical sciences. The method and procedures for section making, observation and counting should be standardized. The authors show the principles and the standardized method based on the case of counting follicles of different developmental stages in the rat ovary, and introduce the recent technical improvement. Taking a particular experiment as an example, this paper discusses the general principles of operation and methods in order to stabilize and improve the quality of microscopic morphological studies.
