摘要: 火棉胶包埋制片法是生物显微研究的一项重要技术,在标本尺寸很大、硬度过高或过低及制作厚切片等场合,具有独特的质量优势。该文根据作者的实践经验,介绍了火棉胶包埋制片法的原理、操作流程和技术要点,展示了该法在上述3种条件下的制片结果和优点。实践表明,如需处理尺寸特别大、硬度很高或很低的标本,或者要求切片既有很大厚度,又要结构精细,应考虑使用本方法。Abstract: The collodion embedding and section making method is one of the important techniques in microscopic research of biology.This method has unique advantages in making high quality sections especially when the specimens are large in scale,the hardness is too high or too low and the production of thick slices.Based on the authors’ practical experience,this article introduces the principle,operation flow and technical points of the cotton wool embedding method,and shows the results and advantages of the method under the above three conditions.Practice shows that is a good method when deals with these particularly large size,high hardness or very low specimens,or slices both large thickness and fine structure.
