酵母醇脱氢酶提纯教学实验的优化探索

An Optimization Study on Teaching Experiment of Yeast Alcohol Dehydrogenase Extraction and Purification

  • 摘要: 针对酵母醇脱氢酶提纯教学实验中存在的问题,为了获得高纯度、高活性、高回收率的酵母醇脱氢酶蛋白,本研究应用单因素实验法对热浸提法、热变性沉淀法、有机溶剂沉淀法分离纯化酶蛋白进行优化,从而提高酵母醇脱氢酶蛋白的提纯效率。结果显示当料液比为1 : 6、粗提温度为25 ℃、粗提时间为1.5 h,热变性温度为65 ℃、时间为15 min,有机溶剂沉淀剂为乙醇、沉淀杂蛋白加入比例为上清液0.5倍,沉淀酶蛋白加入比例为上清液0.45倍时,分离纯化效果最好。通过上述条件的优化,提高了教学实验效果,也有助于锻炼学生的实际动手能力、创新思维和科研能力。

     

    Abstract: There are some problems in the teaching experiment of yeast alcohol dehydrogenase (YADH) extraction and purification. In order to obtain YADH with high purity, high activity, and high recovery, single factor experiment was applied in this study to optimize the separation and purification process of enzyme protein, including hot extraction, thermal denaturation precipitation, and organic solvent precipitation. This will improve the extraction and purification efficiency of YADH. The results showed that the best effect of separation and purification was achieved when the solid-liquid ratio was 1 : 6, the crude extraction temperature was 25 ℃, the crude extraction time was 1.5 h, the thermal denaturation temperature was 65 ℃, the thermal denaturation time was 15 min, the organic solvent precipitant was ethanol, the adding proportion of precipitated impure protein was 0.5 times of the supernatant, and the adding proportion of precipitated enzyme protein was 0.45 times of the supernatant. Through the optimization of the above conditions, the effect of teaching experiment is improved. It is also helpful to exercise students’ practical skills, innovative thinking, and scientific research abilities.
