滑菇菌株对染料吸附脱色作用的综合性实验设计

Comprehensive Experimental Design of Dye Adsorption and Decolorization by Nameko Mushroom Strains

  • 摘要: 为深化本科教学改革,提出在环境微生物实验课程中开设“滑菇菌株对染料吸附脱色作用”的综合性实验。在前序实验完成滑菇菌体培养的基础上,称取成形菌体开展染料脱色实验,通过测定染料溶液吸光度与Zeta电位以及观察菌体表/切面颜色变化等,分析菌株对染料的吸附脱色过程。结果表明,滑菇菌株对不同染料的脱色效果具有明显差异,对10 mg/L BL蓝染料的去除率可达到93.06%。对比不同处理时间下的菌体表/切面图,直观呈现染料在菌体内部的吸附扩散过程,且电负性大的染料与菌体结合更紧密,脱色效果更佳。本实验设计在培养学生基础操作水平的同时,能使学生充分认识到微生物在水处理中的应用,对于培养兼具科研精神和实践能力的高级专业人才有重要意义。

     

    Abstract: In order to deepen the reform of undergraduate teaching, it is proposed to open a comprehensive experiment on “the adsorption and decolorization effect of Pleurotus ostreatus strains on dyes” in the environmental microbiology experiment course. Based on the previous experiment to complete the cultivation of nameko mushroom strains, the formed bacteria were weighed to carry out the dye decolorization experiment. The adsorption and decolorization process of the strain on the dye was analyzed by measuring the absorbance and Zeta potential of the dye solution and observing the color changes of the bacteria surface/cross-section. The results showed that the decolorization effects of nameko mushroom strains on different dyes were significantly different, and the removal rate of 10 mg/L BL blue dye could reach 93.06%. By comparing the bacterial surface/cross-section images at different treatment times, the adsorption and diffusion process of the dye inside the bacteria is intuitively presented. Dyes with high electronegativity are more tightly bound to the bacteria and have better decolorization effects. This experimental design can not only cultivate students' basic operational level, but also enable students to fully understand the application of microorganisms in water treatment. It is of great significance for cultivating senior professionals with both scientific research spirit and practical ability.
