基于SDS诱导果蝇肠炎模型的番石榴多糖提取及活性组分评价综合实验设计

Comprehensive Experimental Design of the Extraction of Guava Polysaccharides and the Evaluation of Active Components Based on the Drosophila Enteritis Model Induced by SDS

  • 摘要: 黑腹果蝇是评价天然产物活性的理想模型之一。本研究构建了一种十二烷基硫酸钠(DSS)诱导的果蝇肠炎模型,用于探究不同提取方法(传统水提醇沉法及复合酶解辅助低共熔溶剂法)及经分离纯化后不同番石榴组分对肠炎的改善作用。结果表明,复合酶解辅助低共熔溶剂法提取的番石榴多糖显著提高了肠炎果蝇存活率,并降低了肠壁通透性。进一步分离纯化后, GPB-1组分在延长果蝇寿命,降低肠壁通透性,恢复肠道长度及提高抗氧化能力方面均优于GPA组分。本研究以果蝇为模型,建立了天然产物活性评价体系,涵盖了多糖提取,分离,纯化以及活性评价的全流程,并结合了生物科学、食品营养学、食品化学、仪器分析等多学科知识,具有较强的综合性。该体系不仅有助于学生掌握天然产物提取工艺、活性评价以及数据分析方法,还为教学提供了一个低成本、伦理友好的实践平台,促进了学生的知识迁移与综合素养的提升。

     

    Abstract: Drosophila melangaster is one of the ideal models for evaluating the activity of natural products. In this study, an SDS-induced intestinal inflammation model of fruit flies was established to explore the improvement effects of different extraction methods (traditional water extraction and alcohol precipitation method and complex enzymatic hydrolysis-assisted deep eutectic solvent method) and different extracts of guava after separation and purification on intestinal inflammation. The results showed that the guava polysaccharides extracted by the complex enzymatic hydrolysis-assisted deep eutectic solvent method significantly increased the survival rate of intestinal inflammation fruit flies and reduced the intestinal wall permeability. After further separation and purification, the GPB-1 component was superior to the GPA component in terms of prolonging the lifespan of fruit flies, reducing intestinal wall permeability, restoring intestinal length and improving antioxidant capacity. This study established a natural product activity evaluation system using fruit flies as a model, covering the entire process of polysaccharide extraction, separation, purification and activity evaluation, and integrating knowledge from multiple disciplines such as biological science, food nutrition, food chemistry and instrumental analysis. It is highly comprehensive. This system not only helps students master natural product extraction techniques, activity evaluation and data analysis methods, but also provides a low-cost and ethically friendly practical platform for teaching, promoting students' knowledge transfer and comprehensive quality improvement.
