曼地亚红豆杉愈伤组织诱导条件探索

Study on the Callus Induction Conditions of Taxus Media

  • 摘要: 该项目以曼地亚红豆杉为试材,研究外植体取材部位、激素种类及浓度组合、光照条件等培养条件对愈伤组织诱导的影响,以找出最适合曼地亚红豆杉愈伤组织生长的培养条件。实验结果发现,最适合诱导曼地亚红豆杉愈伤组织生长的激素组合为0.5 mg/L NAA+0.2 mg/L 6-BA+2 mg/L 2,4-D;最适合外植体为带芽茎段;黑暗培养相比光照培养更有利于愈伤组织生长。该项目的开展可为紫杉醇细胞培养生产体系提供理论指导。

     

    Abstract: Paclitaxel is a highly effective anticancer drug which is mainly extracted from Taxus species,it can be produced by cell culture technology. This study works on the effects of culture conditions such as the types of explants,the types and concentrations of hormones,illumination conditions on callus induction of Taxus media,in order to find out the most suitable culture conditions for Taxus media callus. The results showed that in the experimental conditions,the most suitable hormone combination for Taxus media callus induction was 0.5 mg / L NAA+0.2 mg / L 6-BA+2 mg / L 2,4-D.The most suitable explant was the stem with buds; Dark culture was more conducive to callus growth compared to light culture. This project can provide some references for the production of paclitaxel by cell culture.
