百合细胞悬浮体系构建及多糖含量测定的探索

Construction of Cell Suspension System and the Content Determination of Polysaccharide in Lilium Brownii

  • 摘要: 该文旨在研究百合悬浮体系构建及诱导百合多糖产生的最佳培养方案。首先采用不同取材部位及激素组合来诱导百合愈伤组织形成,将其扩繁进而构建起百合细胞悬浮体系;然后使用不同激素处理来诱导其产生次生代谢产物百合多糖,并通过苯酚硫酸法测定多糖含量。结果发现,百合愈伤组织诱导的最佳取材部位为叶柄,最佳激素配比为6-BA 0.5 mg/L+2,4-D 2.0 mg/L+NAA 0.2 mg/L;百合悬浮细胞培养及诱导百合多糖产生的最佳激素配方为6-BA 1.0 mg/L+2,4-D 2.0 mg/L+NAA 0.2 mg/L,其多糖含量可达到3.8%。该项目可以用于指导百合细胞悬浮体系构建及百合多糖的生产等方面的研究。

     

    Abstract: In this study,the most suitable culture scheme for the construction of Lilium brownii cell suspension system and the inducement of Lilium polysaccharide was researched.Firstly,different parts and hormone treatments were used to induce the callus of Lilium brownii formation,and then the callus was propagated to construct its cell suspension system.After that,the secondary metabolite Lilium polysaccharide was induced by different hormone treatments,and the content of polysaccharide was measured by phenol-sulfuric acid method.It was found that for Lilium brownii callus induction,the best explant was the petiole,and the best hormone formula was 6-BA 0.5 mg/L+2,4-D 2.0 mg/L+NAA 0.2 mg/L.For the culture of Lilium brownii suspension cell and the induction of Lilium polysaccharide,the best hormone formula was 6-BA 1.0 mg/L+2,4-D 2.0 mg/L+NAA 0.2 mg/L,the polysaccharide content was about 3.8%.This project could provide guidance for the construction of Lilium brownii cell suspension system and the production of Lilium polysaccharide.
