基于动态光散射技术对样品溶液质量分析及实验优化

Quality Analysis and Experimental Optimization of Sample Solution Based on Dynamic Light Scattering Technique

  • 摘要: 在生物分子相互作用研究和蛋白纯化等实验中,实验样品溶液质量、蛋白纯度和均一性一直是决定最终实验结果好坏的关键因素。该文利用动态光散射技术,测量溶液样品中分子的自相关函数曲线斜率,解析粒径大小分布,深入分析样品溶液质量的影响因素,优化实验方案。实验结果表明,动态光散射技术为分子生物学实验研究中的条件优化提供了有效的实验依据,大大提高了纯化实验后的蛋白质纯度,改进了生物分子相互作用研究中的实验数据质量。

     

    Abstract: In biomolecular interaction studies and protein purification experiments,the quality of the experimental sample solu-tion,protein purity and homogeneity have been the key factors determining the final experimental results. In this paper,the dynamic light scattering technique is used to measure the slope of the autocorrelation function curve of the molecules in the solution sample,an-alyze the particle size distribution,analyze the influencing factors of the sample solution quality,and optimize the experimental scheme. The experimental results show that the dynamic light scattering technique provided an effective experimental basis for the opti-mization of conditions in molecular biology experimental research,greatly improved the purity of the protein after purification experi-ments and improved the quality of experimental data in the study of biomolecular interactions.
